ABSTRACT

A high-throughput screening machine learning model for mitochondrial function was constructed, and compounds of Aco-niti Lateralis Radix Praeparata were predicted. Deoxyaconitine with the highest score and benzoylmesaconine with the lowest score among the compounds screened by the model were selected for mitochondrial mechanism analysis. Mitochondrial function data were collected from PubChem and Tox21 databases. Random forest and gradient boosted decision tree algorithms were separately used for mo-deling, and ECFP4(extended connectivity fingerprint, up to four bonds) and Mordred descriptors were employed for training, respectively. Cross-validation test was carried out, and balanced accuracy(BA) and overall accuracy were determined to evaluate the performance of different combinations of models and obtain the optimal algorithm and hyperparameters for modeling. The data of Aconiti Lateralis Radix Praeparata compounds in TCMSP database were collected, and after prediction and screening by the constructed high-throughput screening machine learning model, deoxyaconitine and benzoylmesaconine were selected to measure mitochondrial membrane potential, reactive oxygen species(ROS) level and protein expression of B-cell lymphoma 2(Bcl-2), Bcl-2-associated X protein(Bax) and peroxisome proliferator-activated receptor-γ-coactivator 1α(PGC-1α). The results showed that the model constructed using gradient boosted decision tree+Mordred algorithm performed better, with a cross-validation BA of 0.825 and a test set accuracy of 0.811. Deoxyaconitine and benzoylmesaconine changed the ROS level(P<0.001), mitochondrial membrane potential(P<0.001), and protein expression of Bcl-2(P<0.001, P<0.01) and Bax(P<0.001), and deoxyaconitine increased the expression of PGC-1α protein(P<0.01). The high-throughput screening model for mitochondrial function constructed by gradient boosted decision tree+Mordred algorithm was more accurate than that by random forest+ECFP4 algorithm, which could be used to build an algorithm model for subsequent research. Deoxyaconitine and benzoylmesaconine affected mitochondrial function. However, deoxyaconitine with higher score also affected mitochondrial biosynthesis by regulating PGC-1α protein.

ABSTRACT

OBJECTIVE: To investigate the clinical characteristics and related factors of postoperative complications in children with colon polyps treated by endoscopy. METHODS: Clinical data of 379 hospitalized children who underwent electronic colonoscopic polypectomy in Children's Hospital Affiliated to Xi'an Jiaotong University from January 2014 to December 2016 were collected and retrospectively analyzed. RESULTS: Among 379 patients with polypectomy,36 patients(9.1%)had related complications,including fever in 6 patients(1.6%)and bleeding in 30 patients(7.9%);there was no intestinal perforation.Among the 30 cases of bleeding,26 cases(6.9%)were with minor bleeding,and 4 cases(1.1%)were with massive bleeding.The bleeding was successfully stopped by emergency endoscopy,and no additional surgery was performed. Univariate correlation analysis found that the duration of hematochezia,the location of the polyp,being with pedicle or not,the method of resection and postoperative braking were significantly correlated with postoperative complications(P<0.05). Multi-factor Logistic regression analysis found that:the longer the time of hematochezia,the higher the risk of postoperative complications,OR value being 1.08[95%CI:(1.03,2.14),P=0.003]. Patients with non-rectal polyps(including transverse colon,ascending colon and ileocecal)were 18.88 times more likely to have postoperative complications than patients with rectal polyps[95%CI:(2.23,159.74),P=0.007]. The risk of postoperative complications in patients with pedicle was 11.424 times higher than that in patients without pedicle[95%CI:(2.42,53.91),P=0.002]. CONCLUSION: Endoscopic resection of colon polyps in children is safe and effective,and postoperative bleeding is the most common complication. The longer period of hematochezia,the polyp being farther away from the anus,and being with pedicle increases the risk of postoperative complications. The use of titanium clips,strict postoperative braking and softening stool can reduce the occurrence of complications.

ABSTRACT

·AIM: To analyze the curative effect and safety in early stage primary open angle glaucoma (POAG) between simple Ex-PRESS shunt implantation and its combination with biological amniotic membrane implantation. ·METHODS: The clinical data of 53 cases for POAG treated with simple EX-PRESS shunt implantation (22 cases,34 eyes) as control group and its combination with biological amniotic membrane implantation (31 cases,38 eyes) as experimental group were retrospectively analyzed. Intraocular pressure (IOP) preoperatively and postoperatively were statistically evaluated. Filtering bleb and incidences of postoperative complications were observed in our hospital during the period of January of 2015 to April of 2017. ·RESULTS: At each time point after the operation, the mean IOP of the experimental group and the control group decreased distinctly respectively, the difference was statistically significant (P< 0. 05). There was no significant difference on the mean IOP between the experimental group and the control group at 1d and 1wk after operation (P<0.05). The mean IOP in the control group was higher than experimental group at 3 and 6mo (P<0. 05). The rate of functional filtration blebs were different between experimental group and control group during at postoperatively 3mo ( P < 0. 05 ). The complications included shallow anterior chamber, hemorrhage, posterior synechia of the iris, and corneal edema after surgery, and its incidences was not different between two groups (P<0.05). · CONCLUSION: EX - PRESS shunt combined with biological amniotic membrane implantation can effectively and safely reduce intraocular pressure, and improve the condition of filtering bleb after operation, and has long-term effect on IOP.

ABSTRACT

@#AIM: To study the effect of synthetic borneol on transcorneal permeability of dexamethasone. The ocular irritation was also evaluated.<p>METHODS: The freshly excised corneas,whole corneas or de-epithelialized corneas, were immediately mounted in perfusion cells. Dexamethasone containing free-borneol or borneol was added into the deliver chamber respectively in each group. At different time point after addition of solution, 50μL of the receptor solution was removed. The concentration of dexamethasone was determined and the apparent permeability coefficient(Papp)was calculated. Hydration level of cornea was measured and irritation test was observed. <p>RESULTS: The Papp for dexamethasone in whole cornea increased by 2.40 fold in borneol group(<i>P</i><0.05). The Papp for dexamethasone in de-epithelialized cornea did not increased apparently(<i>P</i>>0.05). Synthetic borneol did not increase hydration level and irritation of cornea(<i>P</i>>0.05). <p>CONCLUSION: Synthetic borneol can improve the corneal permeation of lipophilic drug dexamethasone and do not damage corneal tissue.

ABSTRACT

AIM: To investigate the clinical effect of vitamin B12 eye drops for vision fatigue caused by visual display terminals (VDT). METHODS: Totally 50 patients (100 eyes) with vision fatigue caused by VDT were averagely divided into two groups. The control group were treated with normal saline,the treatment group were treated with vitamin B12 eye drops, 3 times per day, one drop each time, continuous for 60d. Accommodative parameters and Schirmer Ⅰtest were measured and analyzed before and after treatment. RESULTS: After treatment, the results of Schirmer Ⅰtest, accommodative amplitude and accommodative facility of the treatment group were higher than those of the control group ( all P CONCLUSION: Vitamin B12 eye drops can lessen symptoms of dry eye, improve accommodative function and treat vision fatigue caused by VDT.

ABSTRACT

<p><b>OBJECTIVE</b>Expression of vimentin in carcinoma cells is a marker for poor prognosis in patients. The aim of this investigation was to assess the influence of vimentin on the characteristics of carcinoma cells.</p><p><b>METHODS</b>The full-length vimentin gene open reading frame (1401 base pairs) was cloned into the plasmid vector pcDNA 3.1 (+), and these vectors were used to stably transfect the human hepatocellular carcinoma HepG2 cell line. Vimentin gene expression was evaluated by RT-PCR and Western blot. Proliferative activity and invasive potential of tumor cells were determined by the CellTiter 96 aqueous one solution cell proliferation assay and BioCoat GFR Matrigel invasion chamber, respectively.</p><p><b>RESULTS</b>DNA sequencing and restriction endonuclease digestion analysis demonstrated that the recombinant vector was correctly cloned. The stable cell line demonstrated a higher vimentin RNA and protein expression. However, both proliferative and invasive abilities of the cells were reduced in vitro ( P < 0.05).</p><p><b>CONCLUSION</b>A recombinant plasmid pcDNA3. 1-VIM is successfully constructed and a carcinoma cell line HepG2-pV highly expressing vimentin is obtained. Recombinant vimentin protein suppresses the proliferative and invasive abilities of HepG2 cells, suggesting that it might decrease malignant phenotype of tumor cells in vitro. This work makes a foundation for further study on the relationship between vimentin and biological phenotype of carcinoma cells.</p>
